Visualizing the fit of the mixed model correlating quartz exposure with over-shift differences (A) and morning levels (B) of caspase-1 ex vivo activation with LPS + ATP in monocytes from participating foundry workers (n = 69 measurements from 40 individuals for the over-shift difference (A), and n = 32 for the morning, 2nd measurement (B)). The parameters quartz exposure, age, BMI, and smoking were included in model (A), and in addition workplace (B) determined by the Akaike information criterion. Caspase-1 enzymatic activity was stimulated by treatment of the blood samples with 10 pg/mL LPS for 3 h, followed by 1 h treatment together with 250 µM ATP and the FAM-YVAD-FMK caspase-1 enzymatic activity probe, before determination of the percent caspase-1 activated monocytes.
